Cover illustration Yeast display of antibody scFv fragments, a powerful technology that gives rapid access to human antibodies, was shown to sample an immune antibody repertoire more fully than phage display, selecting all the specific scFvs identified by phage display and twice as many again of novel scFvs. Expression of scFv on yeast cells is monitored using either the HA or c-myc epitope tags. Binding of the target antigen, HIV-1 gp120, is visualized using a biotinylated mAb to a noncompetitive epitope on gp120 and fluorescent streptavidin. Gp120-binding scFvs are selected by fluorescence activated cell sorting (FACS) of the yeast cells. For further details see Bowley et al. pp. 81-90.
