Cover illustration A human TCR was subjected to phage display to generate high affinity variants that recognise tumour epitope NY-ESO157-165 associated with major histocompatibility complex (pMHC), in the context of human leukocyte antigen (HLA-A*0201). Crystal structures of high affinity TCR/pMHC complexes reveal a native diagonal binding geometry as shown by the domain based multiple alignments of all the mutant models onto the wild-type complex. MHC α1/α2 is red, MHC α3 is brown, β2-microglobulin is cyan,TCRα N-domain is blue,TCRα C-domain is green,TCRβ N-domain is orange and TCRβ C-domain is grey.
